A new, practical synthesis of aryl and heteroaryl N-acylureas has been developed via palladium-catalysed carbonylation of aryl or heteroaryl halides in the presence of urea nucleophiles. A range of reactions illustrating the wide scope of this reaction was carried out under microwave irradiation, using either carbon monoxide gas in a vessel equipped with a gas inlet adapter, or molybdenum hexacarbonyl as the carbon monoxide source in standard microwave vials. The reactions proceeded in good to excellent yields. To illustrate the usefulness of this method a one-step synthesis of the important insecticide diflubenzuron is reported.

A general intermolecular reductive Heck reaction of organohalides with both terminal and internal unactivated aliphatic alkenes has been first realized in high yield with complete anti-Markovnikov selectivity. The challenging vinyl bromides, aryl chlorides, and polysubstituted internal alkenes were first applied. More than 100 remote carbofunctionalized alkyl carboxylic acid derivatives were rapidly synthesized from easily accessible starting materials. The synthesis of drug molecules has further demonstrated the wide synthetic utility of this scalable strategy. Preliminary mechanistic studies are consistent with the proposed catalytic cycle.

Complexation of ca. 1000 ppm Pd(OAc)2 with the ligand HandaPhos (1-1.5:1) leads to a precatalyst that efficiently mediates Sonogashira couplings in aqueous nanomicelles under very mild conditions. Neither copper nor organic solvent is required in the reaction medium, and the product can be isolated directly from the reaction flask, leaving behind a reaction mixture that can be recycled without additional additives.

A new series of calix[4]arene supported N-heterocyclic carbene palladium complexes were developed and fully characterized. A mono-substituted calix[4]arene was prepared through conventional procedures, following with the attachment of imidazolyl derivative groups to compose the precursors of novel NHCs ligands. Undergoing the alkylation with n-butylbromide and corresponding metallation with palladium and pyridine, original complexes were obtained. The green generation of X+ (X = Br, I) using hydrogen peroxide in aqueous acetic acid allows access to aromatic heterocyclic halides in yields and purities comparable to syntheses employing N-bromosuccinimide. In activated and unsubstituted thiophene rings, regioselectivity is quantitative for positions alpha to the sulfur; pyrroles also give quantitative reactions, at least initially. Deactivated rings, including furans and thiazoles, as well as thiophenes with strongly electron-withdrawing groups showed little to no reactivity under the conditions investigated. The reaction shows remarkable functional group tolerance (to alcohol, nitro, alkyl, halo, and carbonyl groups), as shown through reaction with substituted phenols. In all bromination reactions, reaction yields and regiochemistry were very similar to reactions involving N-bromosuccinimide in tetrahydrofuran solvent.

Heterogeneous palladium catalyst constructed with cross-linked hyperbranched poly(phenylacetylene) as polymer support: A reusable highly active ppm-level catalyst for multiple cross-coupling reactions

We demonstrate in this work a unique strategy for the synthesis of heterogeneous Pd catalysts with the use of a hyperbranched poly(phenylacetylene) containing pendant alkyne groups as the cross-linkable polymer substrate. It utilizes the dual functions of Pd-based catalysts in catalyzing both alkyne polymerization/oligomerization and coupling reactions. In the synthesis, a homogeneous Pd(II) catalyst catalyzes the cross-linking of the hyperbranched polymer and simultaneously encapsulates itself into the crosslinked polymer matrix, rendering the heterogeneous catalyst at high yield and high percentage of Pd encapsulation. Three homogeneous catalysts having different ligands (triphenylphosphine, a diphosphine ligand, and a diimine ligand) have been examined for the cross-linking encapsulation and the resulting heterogeneous catalysts have been evaluated for their catalytic performance in coupling reactions. Among the various heterogeneous catalysts obtained, a triphenylphosphine-ligated catalyst, HBPPA-Pd-2, appears to be the optimum one. It shows high activity in catalyzing the Suzuki-Miyaura reactions, the Mizoroki-Heck reactions, and the allylic arylation reactions under air with the Pd loading at as low as mol ppm or even mol ppb levels relative to the reactants. Meanwhile, it facilitates the Suzuki-Miyaura reactions of challenging less reactive aryl chlorides/heteroaryl bromides and the Mizoroki-Heck reactions of aryl bromides as reactants. In addition, it behaves truly as a heterogeneous catalyst with high reusability and low catalyst leaching during the reactions.

Synthesis, binding assays, cytotoxic activity and docking studies of benzimidazole and benzothiophene derivatives with selective affinity for the CB2 cannabinoid receptor

Herein we report the design, synthesis, bioinformatic and biological studies of benzimidazole and benzothiophene derivatives as new cannabinoid receptor ligands. To test the hypothesis that the lack of a hydrogen bond interaction between benzimidazole and benzothiophene derivatives with Lys192 reduces their affinity for CB1 receptors (as we previously reported) and leads to CB2 selectivity, most of the tested compounds do not exhibit hydrogen bond acceptors. All compounds displayed mostly CB2 selectivity, although this was more pronounced in the benzimidazoles derivatives. Furthermore, docking assays revealed a ?-cation interaction with Lys109 which could play a key role for the CB2 selectivity index. The series displayed low toxicity on five different cell lines. Derivative 8f presented the best binding profile (Ki = 0.08 muM), high selectivity index (KiCB1/KiCB2) and a low citoxicity. Interestingly, in cell viability experiments, using HL-60 cells (expressing exclusively CB2 receptors), all synthesised compounds were shown to be cytotoxic, suggesting that a CB2 agonist response may be involved.
