The stereoselective synthesis of syn-beta-fluoroaziridine building blocks via chiral aryl iodide-catalyzed fluorination of allylic amines is reported. The method employs HF-pyridine as a nucleophilic fluoride source together with mCPBA as a stoichiometric oxidant, and affords access to arylethylamine derivatives featuring fluorine-containing stereocenters in high diastereo- and enantioselectivity. Catalyst-controlled diastereoselectivity in the fluorination of chiral allylic amines enabled the preparation of highly enantioenriched 1,3-difluoro-2-amines bearing three contiguous stereocenters. The enantioselective catalytic method was applied successfully to other classes of multifunctional alkene substrates to afford anti-beta-fluoropyrrolidines, as well as a variety of 1,2-oxyfluorinated products.

The syntheses and antiplasmodial activities of various substituted aminoquinolines coupled to an adamantane carrier are described. The compounds exhibited pronounced in vitro and in vivo activity against Plasmodium berghei in the Thompson test. Tethering a fluorine atom to the aminoquinoline C(3) position afforded fluoroaminoquinolines that act as intrahepatocytic parasite inhibitors, with compound 25 having an IC50 = 0.31 muM and reducing the liver load in mice by up to 92% at 80 mg/kg dose. Screening our peroxides as inhibitors of liver stage infection revealed that the tetraoxane pharmacophore itself is also an excellent liver stage P. berghei inhibitor (78: IC50 = 0.33 muM). Up to 91% reduction of the parasite liver load in mice was achieved at 100 mg/kg. Examination of tetraoxane 78 against the transgenic 3D7 strain expressing luciferase under a gametocyte-specific promoter revealed its activity against stage IV-V Plasmodium falciparum gametocytes (IC50 = 1.16 ± 0.37 muM). To the best of our knowledge, compounds 25 and 78 are the first examples of either an 4-aminoquinoline or a tetraoxane liver stage inhibitors.

The HOMO and lowest unoccupied molecular orbital (LUMO) in PBDTBzT-DTffBT are decreased by 0.11 and 0.13 eV, respectively, which should be attributed to the contribution of the electron-withdrawing group benzene. Polymer solar cells (PSCs) based on PBDTBzT-DTffBT and [6,6]-phenyl-C61-butyric acid methyl ester (PC61BM) exhibit a maximum power conversion efficiency (PCE) of 7.30% with a large open-circuit voltage of 0.90 V under AM 1.5G illumination (100 mW/cm2). The PCE is 36% higher than that of the PSCs derived from PBDTT-DTffBT. These findings provide a new approach to design high-performance conjugated polymers for efficient solution-processed PSCs.
