The respective conjugate bases, 3–O and 3–S, represent benzofuran and benzothiophene derivatives, respectively, and thus are aromatic. The main question addressed in this paper is whether this aromaticity has the effect of enhancing or lowering intrinsic barriers to proton transfer. These intrinsic barriers were either determined from Bronsted plots for the reactions with amines or calculated on the basis of the Marcus equation for the reaction with OH-; they were found to be lower for the more highly aromatic benzothiophene derivative, indicating that aromaticity lowers the intrinsic barrier. It is shown that the reduction in the intrinsic barrier is not an artifact of other factors such as inductive, steric, resonance, polarizability, and pi-donor effects, although these factors affect the intrinsic barriers in a major way. Our results imply that aromatic stabilization of the transition state is ahead of proton transfer; this contrasts with simple resonance effects, which typically lag behind proton transfer at the transition state, thereby increasing intrinsic barriers.

The abilities of three Pseudomonas strains to transform each of the six isomers of methylbenzothiophene was studied. These compounds would not support growth but were biotransformed in a cell suspension of one of the isolates and in cultures of the other two isolates while they grew on 1- methylnaphthalene or glucose. Sulfur-containing metabolites were identified by GC/MS and GC/FTIR, and when possible, by comparison with authentic standards. Sulfoxides and sulfones were frequently detected and were the most abundant products from 2- and 3-methylbenzothiophene. Two of the isolates oxidized the methyl groups to carboxylic acids which were abundant products from 3-, 5-, and 6-methylbenzothiophenes. Benzothiophenemethanols were often detected, but in very low amounts. 2,3-Diones were observed as metabolites of benzothiophene and those methylbenzothiophenes with a methyl group on the benzene ring. Isomers of tolyl methyl sulfoxide were observed as ring cleavage products from 6- and 7-methylbenzothiophene.

Previous works of our group have dealt with the synthesis of 1-(aryl)-3-[4-(aryl)piperazin-1-yl]propane derivatives in the search for new and efficient antidepressants with a dual mode of action: serotonin reuptake inhibition and 5-HT1A receptor afinity [1-4]. From these studies we concluded that the 3-[4-(aryl)piperazin-1-yl]-l-(benzo[b]thiophen-3-yl)propane derivatives led to the best results. The continuation of this research project required the preparation of some new 3-acyl-5-substituted benzo[b]thiophenes with a wide variety of substituents at the 5 position, ranging from nitro to hydroxyl derivatives. To obtain these derivatives we acylated the corresponding 5-substituted benzo[b]thiophenes when it was possible.
