SGC-GAK-1 (1) is a potent, selective, cell-active chemical probe for cyclin G-associated kinase (GAK). However, 1 was rapidly metabolized in mouse liver microsomes by cytochrome P450-mediated oxidation, displaying rapid clearance in liver microsomes and in mice, which limited its utility in in vivo studies. Chemical modifications of 1 that improved metabolic stability, generally resulted in decreased GAK potency. The best analog in terms of GAK activity in cells was 6-bromo-N-(1H-indazol-6-yl)quinolin-4-amine (35) (IC50 = 1.4 muM), showing improved stability in liver microsomes while still maintaining a narrow spectrum activity across the kinome. As an alternative to scaold modifications we also explored the use of the broad-spectrum cytochrome P450 inhibitor 1-aminobenzotriazole (ABT) to decrease intrinsic clearance of aminoquinoline GAK inhibitors. Taken together, these approaches point towards the development of an in vivo chemical probe for the dark kinase GAK.

Here, we describe the straightforward synthesis of chiral amines by combining the atom-economic and environmentally friendly hydroamination of alkynes with an enantioselective hydrogenation of in situ generated imines by using inexpensive hydrogen. By following this novel approach, a wide range of terminal alkynes can be reductively hydroaminated with primary amines including alkyl-, and arylalkynes as well as aryl and heteroaryl amines. Excellent yields and selectivities up to 94 % ee and 96 % isolated yield were obtained.

Recent studies conducted at our laboratory have shown that sediments contaminated with complex mixtures of polycyclic aromatic hydrocarbons (PAHs) can exhibit enhanced toxicity (lethality) to a variety of aquatic species when the samples are tested under ultraviolet (UV) light designed to mimic the wavelengths present in sunlight. However, because these contaminated sediments can contain literally thousands of chemicals, it is difficult to use conventional analytical techniques to identify those compounds responsible for photo-induced toxicity. The purpose of this study was to adapt existing toxicity identification evaluation methods to attempt to identify those compounds contributing to the phototoxicity observed in our sediment samples. Pore water obtained from sediments collected near an oil refinery discharge was toxic to Lumbriculus variegatus following exposure to UV light, while organisms exposed to the same pore water, but without subsequent UV treatment, showed no toxic effect. Solid-phase extraction disks and high-performance liquid chromatography were used, in conjunction with toxicity tests with L. variegatus, to extract and fractionate phototoxic chemicals from the pore water. Phototoxic fractions analyzed by gas chromatography-mass spectrometry revealed the presence of a number of aliphatic hydrocarbons, substituted PAHs, and PAHs containing heteroatoms. Chemicals were screened for their phototoxic potential based on empirical data and predictive models. A refined list of PAHs was then evaluated on the basis of their phototoxic potency as defined by a recently developed quantitative structure-activity relationship model and estimates of their bioaccumulation potential. Based on the model predictions of potency and bioaccumulation, nine likely phototoxic chemicals were identified.

Previous works of our group have dealt with the synthesis of 1-(aryl)-3-[4-(aryl)piperazin-1-yl]propane derivatives in the search for new and efficient antidepressants with a dual mode of action: serotonin reuptake inhibition and 5-HT1A receptor afinity [1-4]. From these studies we concluded that the 3-[4-(aryl)piperazin-1-yl]-l-(benzo[b]thiophen-3-yl)propane derivatives led to the best results. The continuation of this research project required the preparation of some new 3-acyl-5-substituted benzo[b]thiophenes with a wide variety of substituents at the 5 position, ranging from nitro to hydroxyl derivatives. To obtain these derivatives we acylated the corresponding 5-substituted benzo[b]thiophenes when it was possible.
