One-Pot Protocol to Synthesize 2-Aminophenols from Anilines via Palladium-Catalyzed C-H Acetoxylation

This paper describes a facile one-pot protocol to synthesize 2-aminophenol derivatives via a palladium-catalyzed C-H acetoxylation strategy with 5-nitropyrimidine as a directing group (DG), which can be easily preinstalled and readily removed under mild condition after the coupling. In addition, the transformation is operationally simple, has high functional group tolerance, and is amenable to gram-scale. Moreover, several examples were shown that introduction/removal of 5-nitropyrimidine and the C-H oxylation sequence could be integrated in one pot.

The abilities of three Pseudomonas strains to transform each of the six isomers of methylbenzothiophene was studied. These compounds would not support growth but were biotransformed in a cell suspension of one of the isolates and in cultures of the other two isolates while they grew on 1- methylnaphthalene or glucose. Sulfur-containing metabolites were identified by GC/MS and GC/FTIR, and when possible, by comparison with authentic standards. Sulfoxides and sulfones were frequently detected and were the most abundant products from 2- and 3-methylbenzothiophene. Two of the isolates oxidized the methyl groups to carboxylic acids which were abundant products from 3-, 5-, and 6-methylbenzothiophenes. Benzothiophenemethanols were often detected, but in very low amounts. 2,3-Diones were observed as metabolites of benzothiophene and those methylbenzothiophenes with a methyl group on the benzene ring. Isomers of tolyl methyl sulfoxide were observed as ring cleavage products from 6- and 7-methylbenzothiophene.

Benzo[b]thiophene-6-carboxamide 1,1-dioxides: Inhibitors of human cancer cell growth at nanomolar concentrations

Benzo[b]thiophenesulfonamide 1,1-dioxide derivatives (BTS) were described as candidate antineoplastic drugs. In the hope of finding new compounds with improved antitumour activity and reduced toxicity, we have designed and synthesized a small series of benzo[b]thiophene-6-carboxamide 1,1-dioxide derivatives (BTC) structurally related with the best reported BTS. Growth inhibition of HTB-54, CCRF-CEM and HeLa tumour cells lines at nanomolar concentrations was exhibited by some of the BTC. Hydrophobic substituents on the carboxamide group increased cytotoxicity but substitution by a hydroxy group diminished it, thus pointing to the electronic density on benzo[b]thiophene nucleus as a determinant factor. The process of cell death induced by BTC derivatives was further analyzed in CCRF-CEM cells, where these compounds induced apoptosis in a time and dose-dependent manner and cell cycle arrest at S phase. BTC derivatives also induced a significant increase in intracellular ROS levels in this cell line. Previous treatment of the cells with the antioxidant N-acetyl-cysteine abrogated the induction of apoptosis by BTC indicating that ROS generation is a previous event required to trigger the BTC induced apoptotic process.

However, shifting the wavelengths for bistable photoswitching beyond 600 nm is still a great challenge without involving secondary processes such as two-photon absorption or sensitization. We present a simple hemithioindigo photoswitch that can efficiently be photoisomerized using green and red light while maintaining a high thermal barrier of the metastable state. This highly sought after properties allow for selective switching in a mixture of hemithioindigo dyes. In addition, protonation can be used as second independent input altering the light response of the switch and allowing construction of advanced molecular digital information processing devices. This is demonstrated by realizing a broad variety of logical operations covering combinational and sequential logic behavior. By making use of the protonation-induced loss of thermal bistability, a high security keypad lock could be realized, which distinguishes the sequences of three different inputs and additionally erases its unlocked state after a short time.

C17,20-lyase inhibitors I. Structure-based de novo design and SAR study of C17,20-lyase inhibitors.

Novel nonsteroidal C(17,20)-lyase inhibitors were synthesized using de novo design based on its substrate, 17 alpha-hydroxypregnenolone, and several compounds exhibited potent C(17,20)-lyase inhibition. However, in vivo activities were found to be short-lasting, and in order to improve the duration of action, a series of benzothiophene derivatives were evaluated. As a result, compounds 9h, (S)-9i, and 9k with nanomolar enzyme inhibition (IC(50)=4-9 nM) and 9e (IC(50)=27 nM) were identified to have powerful in vivo efficacy with extended duration of action. The key structural determinants for the in vivo efficacy were demonstrated to be the 5-fluoro group on the benzothiophene ring and the 4-imidazolyl moiety. Superimposition of 9k and 17 alpha-hydroxypregnenolone demonstrated their structural similarity and enabled rationalization of the pharmacological results. In addition, selected compounds were also identified to be potent inhibitors of human enzyme with IC(50) values of 20-30 nM.
