From these results, we found the following three differences between perhydrocyclopentenes and perfluorocyclopentenes. (1) For the closed-ring isomers, the absorption bands of the perhydrocyclopentenes (around 450 nm) shifted to a shorter wavelength than those of the perfluorocyclopentenes (around 530 nm) because of the acceptor nature of the perfluorocyclopentene moiety. (2) The cyclization/cycloreversion quantum yields of the perhydrocyclopentenes were slightly larger than those of the perfluorocyclopentenes in hexane. (3) Interestingly, the fluorescence of both the open- and closed-ring isomers was observed only for the perhydrocyclopentenes, and the lifetimes of the closed-ring isomers were estimated to be <30 ps. The fluorescence of the closed-ring isomers of the perhydrocyclopentenes was mainly due to the suppression of nonradiative deactivation from the excited state directly to the ground state compared with the perfluorocyclopentenes. Thrombosis is a pathological coagulation process and can lead to many serious thrombotic diseases. Here, we report a novel potent antithrombotic compound (6k) based on isosteviol with anticoagulant and antiplatelet activities. 6k selectively inhibited FXa (Ki = 0.015 muM) against a panel of serine proteases and showed excellent anticoagulant activity (significant prolongation of ex vivo PT and aPTT over the vehicle, p < 0.01). 6k also significantly inhibited ADP-induced platelet aggregation in rats relative to the vehicle (p < 0.01). Furthermore, 6k exhibited potent ex vivo and in vivo antithrombotic activity in rats relative to the vehicle (p < 0.01 and p < 0.0001, respectively). Novel structure 6k, with potent antithrombotic activity, is expected to lead a promising approach for the development of antithrombotic agents. A series of 2,4-dioxo-thieno[2,3-d], [3,2-d] and [3,4-d]pyrimidin-1-acetic acids (2) with a benzyl moiety at the N-3 position were prepared and tested in vitro for aldose reductase inhibitory activity against partially purified enzyme from rat lens. Some of these compounds were also evaluated for inhibition of sorbitol accumulation in the sciatic nerve or lens of streptozotocin-induced diabetic rats in vivo. Among the synthesized compounds, several showed potent aldose reductase inhibitory activity with IC50s in the 10-8 M range. Particularly, the potencies of non-substituted thieno- (2a and 2aa), 5-methylthieno- (2c), 5,6-dimethylthieno- (2g), 6-isopropylthieno- (2j and 2k), 6-chlorothienopyrimidine (2q) and benzothienopyrimidine (2ac) analogs were approximately equipotent ot FK-366 (1A) and Ponalrestat (1B) as references. Although most compounds were inactive in vivo, 2 compounds, 2k and 2q, possessed moderate in vivo activity.
