Optical properties of dibenzo[d,d?]thieno[3,2-b;4,5-b?] dithiophene monocrystals: The effect of intermolecular interactions

The polarized UV-visible absorption spectra of dibenzo[d,d?]thieno[3, 2-b;4,5-b?]dithiophene single crystals are reported and interpreted to definitively attribute the observed bands and their polarizations. The results provide information on the intermolecular interactions and on the aggregation in the condensed phase, which can be of either herringbone- or H-type, depending on the electronic transition taken into considerations, with consequences on the order and polarization of the absorption bands. A relatively easy method is also discussed to obtain information on the structural/morphological properties of different types of samples, including thin films, which have been recently proposed for high-performance organic film-effect transistors for their high ionization potential and photostability.

A novel family of photochromic amorphous molecular materials containing a dithienylethene moiety, 1-{5-[4-(di-p-tolylamino)phenyl]-2-methylthiophen-3-yl}-2- (2,5-dimethylthiophen-3-yl)-3,3,4,4,5,5-hexafluorocyclopentene, 1-{5-[4-(di-p-tolylamino)phenyl]-2-methylthiophen-3-yl}-2- (2-methylbenzo[b]thiophen-3-yl)-3, 3,4,4,5,5-hexafluorocyclopentene and 1,2-bis{5-[4-(di-p-tolylamino)phenyl]-2-methylthiophen-3-yl}-3, 3,4,4,5,5-hexafluorocyclopentene, have been designed and synthesised. These compounds, together with their photocyclised products, were found to readily form amorphous glasses with well-defined glass transition temperatures and to undergo photochromism as amorphous films as well as in solution. These compounds are characterised by high quantum yields for the photocyclisation reactions, very low quantum yields for the reverse ring-opening reactions and, hence, almost 100% fractions of the photocyclised form at the photostationary state in solution. These results suggest that the anti-parallel conformer is more populated than the parallel conformer. Optical dichroism was induced by irradiation of coloured films of the photocyclised compounds with linearly polarised red light, and dual image formation at the same location was realised by utilising this phenomenon.

The high cost associated with palladium, combined with several limitations of both palladium- and copper-catalyzed processes, prompted us to develop an alternative method. Thus, the nickel-catalyzed borylation of aryl and heteroaryl halides and pseudohalides using tetrahydroxydiboron (BBA) has been formulated. The reaction proved to be widely functional group tolerant and applicable to a number of heterocyclic systems. To the best of our knowledge, the examples presented here represent the only effective Ni-catalyzed Miyaura borylations conducted at room temperature.

Raloxifene,[2-(4-hydroxyphenyl)-6-hydroxybenzo[b]thien-3-yl][4-[2-(1- piperidinyl)ethoxy]phenyl]-methanone hydrochloride (2), is representative of a class of compounds known as selective estrogen receptor modulators (SERMs) that possess estrogen agonist-like actions on bone tissues and serum lipids while displaying potent estrogen antagonist properties in the breast and uterus. As part of ongoing SAR studies with raloxifene, we found that replacement of the carbonyl group with oxygen ([6-hydroxy-3-[4-[2-(1- piperidinyl)ethoxy]phenoxy]-2-(4-hydroxyphenyl)]benzo[b]thiophene hydrochloride, 4c) resulted in a substantial (10-fold) increase in estrogen antagonist potency relative to raloxifene in an in vitro estrogen dependent cell proliferation assay (IC50 = 0.05 nM) in which human breast cancer cells (MCF-7) were utilized. In vivo, 4c potently inhibited the uterine proliferative response to exogenous estrogen in immature rats following both sc and oral dosing (ED50 of 0.006 and 0.25 mg/kg, respectively). In ovariectomized aged rats, 4c produced a significant maximal decrease (45%) in total cholesterol at 1.0 mg/kg (po) and showed a protective effect on bone relative to controls with maximal efficacy at 1.0 mg/kg (po). These data identify 4c as a novel SERM with greater potency to antagonize estrogen in uterine tissue and in human mammary cancer cells compared to raloxifene, tamoxifen or ICI-182,780.
